Background

E3 ubiquitin-protein ligase involved in different protein quality control pathways in the cytoplasm and nucleus . Mainly acts as a ubiquitin chain elongator that extends pre-ubiquitinated substrates . Component of the N-end rule pathway: ubiquitinates proteins bearing specific N-terminal residues that are destabilizing according to the N-end rule, leading to their degradation . Recognizes type-1 N-degrons, containing positively charged amino acids (Arg, Lys and His) .

Western blot analysis of UBR5 expression in zebrafish (A) whole cell lysates. (Predicted band size: 309 kD; Observed band size: 309 kD)

Immunohistochemical analysis of UBR5 staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
