Product Details
Intended Use
For epitope retrieval prior to immunohistochemical staining.
Detection Principle
Tissues or cells fixed with paraformaldehyde, formaldehyde or other aldehydes develop protein cross-links that mask antigen epitopes and weaken immuno-staining signals.
Heating tissue sections in this buffer restores masked epitopes, breaks cross-linked peptide chains without destroying epitope structures. It enhances specific staining intensity, minimizes non-specific background, and reduces false positive and false negative results.
Main Components
EDTA, Tris, surfactant, etc.
Other required supplies not included:
Wash buffer, deparaffinization solution, hematoxylin, IHC chromogenic substrates, graduated cylinders, plastic humid chambers, slide racks, stainless steel pots or pressure cookers.
Storage Conditions and Shelf Life
Storage: Room temperature (2–30 °C); shelf life: 18 months.
Use within 3 months after opening.
Production date and expiry date: see product label.
Sample Requirements
Formalin-fixed, paraffin-embedded tissue sections.
Detection Procedure
Ready-to-use without dilution; apply as required.
1.Automated IHC staining system: Pour the buffer into the designated tank of the automated tissue stainer.
2.Manual IHC staining: Perform epitope retrieval following standard protocols.
Result Interpretation
Interpretation depends on tissue type and experimental purposes.
Note: For clinical IHC testing, results must be interpreted by qualified pathologists.

Influencing Factors
1.Operators shall receive professional training and strictly follow SOPs.
2.All measuring tools and equipment must be calibrated to avoid testing errors.
Limitations
1.Standardized testing relies on trained personnel and certified laboratories to reduce operational deviations.
2.Optimal heat retrieval time varies due to inconsistent tissue fixation; users must optimize conditions via preliminary trials.
Performance Indicators
1.Appearance: Clear product labeling; colorless, homogeneous liquid.
2.Net volume: Not less than labeled volume.
3.pH Compliance: pH range 8.5–9.5 (measured at 25±1°C).
4.Intra-batch Repeatability: pH values of single batch fall within 8.5–9.5 (25±1°C).
5.Inter-batch Repeatability: pH values across batches remain 8.5–9.5 (25±1°C).
Precautions
1.This buffer is exclusively for epitope retrieval of FFPE tissue sections before IHC staining.
2.Read the full manual before starting experiments.
3.Use before the expiry date.
4.Only for professionally trained or experienced laboratory staff.
5.Over-retrieval causes abnormal staining; insufficient retrieval leads to weak signals.
6.Conduct pre-experiments to confirm optimal retrieval conditions; use adhesive anti-drop slides.
7.Avoid contact with eyes and mucous membranes; flush thoroughly with plenty of clean water upon contact.
8.Wear latex gloves, masks, goggles and lab coats for personal protection.
9.Dispose of all experimental waste in compliance with Regulations on the Administration of Medical Wastes.
