Background
DNA- and RNA binding protein, involved in several nuclear processes . Binds the conventional octamer sequence in double-stranded DNA . Also binds single-stranded DNA and RNA at a site independent of the duplex site . Involved in pre-mRNA splicing, probably as a heterodimer with SFPQ . Interacts with U5 snRNA, probably by binding to a purine-rich sequence located on the 3' side of U5 snRNA stem 1b . Together with PSPC1, required for the formation of nuclear paraspeckles . The SFPQ-NONO heteromer associated with MATR3 may play a role in nuclear retention of defective RNAs . The SFPQ-NONO heteromer may be involved in DNA unwinding by modulating the function of topoisomerase I/TOP1 .

Western blot analysis of p54nrb expression in K562 (A), Hela (B), MCF7 (C) whole cell lysates. (Predicted band size: 54 kD; Observed band size: 54 kD)

Immunofluorescent analysis of p54nrb staining in MDAMB231 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a AREX® Fluor 488 -conjugated secondary antibody (green) in PBS at room temperature in the dark. Phalloidin - AREX® Fluor 594 was used to stain Actin filaments (red). DAPI was used to stain the cell nuclei (blue).
