Background

Receptor for IL17A and IL17F, major effector cytokines of innate and adaptive immune system involved in antimicrobial host defense and maintenance of tissue integrity. Receptor for IL17A . Receptor for IL17F . Binds to IL17A with higher affinity than to IL17F . Binds IL17A and IL17F homodimers as part of a heterodimeric complex with IL17RC . Also binds heterodimers formed by IL17A and IL17F as part of a heterodimeric complex with IL17RC . Cytokine binding triggers homotypic interaction of IL17RA and IL17RC chains with TRAF3IP2 adapter, leading to TRAF6-mediated activation of NF-kappa-B and MAPkinase pathways, ultimately resulting in transcriptional activation of cytokines, chemokines, antimicrobial peptides and matrix metalloproteinases, with potential strong immune inflammation .

Western blot analysis of IL-17RA expression in rat spleen (A), mouse lung (B), MCF7 (C) whole cell lysates. (Predicted band size: 96 kD; Observed band size: 150 kD)

Immunofluorescent analysis of IL-17RA staining in MCF7 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a AREX® Fluor 488 -conjugated secondary antibody (green) in PBS at room temperature in the dark. Phalloidin - AREX® Fluor 594 was used to stain Actin filaments (red). DAPI was used to stain the cell nuclei (blue).

Storage

Store at 4°C short term. For long term storage, store at -20°C, avoiding freeze/thaw cycles.
