Background

Core component of nucleosome. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human endometrial cancer tissue labelling Histone 3.1 with ARA799 at 1:800. Heat mediated antigen retrieval was performed using Tris/EDTA buffer pH 9.0.

Overlay histogram showing Hela cells stained with ARA799 (Red). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% TritonX-100 for 15 min. The cells were then incubated in the antibody (ARA799, 1:200 dilution) in 1x PBS/1% BSA for 30 min at room temperature. The secondary antibody used was a Goat Anti-Rabbit Alexa Fluor<sup>®</sup> 488 (IgG H+L) at 1:2,000 dilution for 20 min at room temperature. Unlabelled sample (Black) was used as a control.

Histone 3.1 was immunoprecipitated from 0.4mg of Hela whole cell lysate with ARA799 at 1:100 dilution.
