Background

Receptor for the chemotactic and inflammatory peptide anaphylatoxin C5a, stimulating chemotaxis, granule enzyme release, intracellular calcium release and superoxide anion production . Also acts as a receptor for dearginated forms of C3a, C4a and C5a anaphylatoxin peptides (ASP/C3adesArg, C4adesArg and C5adesArg, respectively) . Ligand binding causes a conformation change that triggers signaling via guanine nucleotide-binding proteins (G proteins) and modulates the activity of downstream effectors, such as adenylate cyclase . C5AR1 is coupled to G(i)/G(o) (GNAI1 or GNAO1) G alpha proteins and mediates inhibition of adenylate cyclase .

Western blot analysis of GPR77 expression in K562 (A), THP1 (B), MOLT4 (C) whole cell lysates. (Predicted band size: 36 kD; Observed band size: 36 kD kD)

Immunohistochemical analysis of GPR77 staining in human liver cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
