Background

Receptor for the cytotoxic ligand TNFSF10/TRAIL . The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis . Promotes the activation of NF-kappa-B .

Western blot analysis of DR4 expression in A375 (A), A2780 (B), rat spleen (C) whole cell lysates. (Predicted band size: 50 kD; Observed band size: 56 kD)

Immunohistochemical analysis of DR4 Antibody staining in human lung cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
