Background

DNMT3B is a DNA-(cytosine C5)-methyltransferase that is responsible for establishing the methylation patterns in cooperation with DNMT3A through the de novo methylation pathway. It is essential for the establishment of DNA methylation patterns during development. DNMT3B contains a C-terminal catalytic domain and an N-terminal regulatory domain, which are involved in targeting chromatin to regulate its function (PMID: 31547729). DNMT3B has 8 isoforms with the molecular mass of 80-96 kDa.

Western blot analysis of DNMT3B expression in A431 (A) whole cell lysates. (Predicted band size: 96 kD; Observed band size: 96 kD)

Immunofluorescent analysis of DNMT3B staining in A431 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with an AREX Flour 488-conjugated secondary antibody (green) in PBS at room temperature in the dark. Phalloidin - AREX Flour was used to stain Actin filaments (red). DAPI was used to stain the cell nuclei (blue).

Storage

Store at 4°C short term. For long term storage, store at -20°C, avoiding freeze/thaw cycles.
