Background

Thiol protease that acts as a major effector caspase involved in the execution phase of apoptosis . Following cleavage and activation by initiator caspases (CASP8, CASP9 and/or CASP10), mediates execution of apoptosis by catalyzing cleavage of many proteins . At the onset of apoptosis, it proteolytically cleaves poly(ADP-ribose) polymerase PARP1 at a '216-Asp-|-Gly-217' bond . Cleaves and activates sterol regulatory element binding proteins (SREBPs) between the basic helix-loop-helix leucine zipper domain and the membrane attachment domain . Cleaves and activates caspase-6, -7 and -9 (CASP6, CASP7 and CASP9, respectively) . Cleaves and inactivates interleukin-18 (IL18) . Involved in the cleavage of huntingtin . Triggers cell adhesion in sympathetic neurons through RET cleavage .

Western blot analysis of Caspase 3 expression in SGC7901 (A), mouse lung (B), Hela (C) whole cell lysates. (Predicted band size: 31 kD; Observed band size: 35 kD)

Immunofluorescent analysis of Caspase 3 staining in KYSE30 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a AREX® Fluor 488 -conjugated secondary antibody (green) in PBS at room temperature in the dark. Phalloidin - AREX® Fluor 594 was used to stain Actin filaments (red). DAPI was used to stain the cell nuclei (blue).
