Background

Calcium-regulated non-lysosomal thiol-protease which catalyzes limited proteolysis of substrates involved in cytoskeletal remodeling and signal transduction . Proteolytically cleaves CTBP1 at 'Asn-375', 'Gly-387' and 'His-409' . Cleaves and activates caspase-7 (CASP7) .

Western blot analysis of Calpain 1 expression in Jurkat (A), K562 (B), MCF7 (C), PC3 (D) whole cell lysates. (Predicted band size: 82 kD; Observed band size: 80 kD kD)

Immunohistochemical analysis of Calpain 1 staining in human cervical cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
