Background

Plays a role in the mitochondrial apoptotic process . Under normal conditions, BAX is largely cytosolic via constant retrotranslocation from mitochondria to the cytosol mediated by BCL2L1/Bcl-xL, which avoids accumulation of toxic BAX levels at the mitochondrial outer membrane (MOM) . Under stress conditions, undergoes a conformation change that causes translocation to the mitochondrion membrane, leading to the release of cytochrome c that then triggers apoptosis . Promotes activation of CASP3, and thereby apoptosis .

Western blot analysis of BAX expression in Hela (A), C2C12 (B), C6 (C), HepG2 (D), MCF7 (E), Ramos (F), Raji (G), HEK293 (H), HEK293-6e (I) whole cell lysates. (Predicted band size: 21 kD; Observed band size: 50 kD kD)

Immunohistochemical analysis of BAX staining in human cervical cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
