Background

Integral membrane protein that functions as a NADPH-dependent ferric-chelate reductase, using NADPH from one side of the membrane to reduce a Fe(3+) chelate that is bound on the other side of the membrane. Mediates sequential transmembrane electron transfer from NADPH to FAD and onto heme, and finally to the Fe(3+) chelate chelate that is bound on the other side of the membrane. Mediates sequential transmembrane electron transfer from NADPH to FAD and onto heme, and finally to the Fe(3+) chelate . Can also reduce Cu(2+) to Cu(1+) . Plays a role in systemic metabolic homeostasis, integrating inflammatory and metabolic responses . Associated with obesity and insulin-resistance . Involved in inflammatory arthritis, through the regulation of inflammatory cytokines .

Western blot analysis of STEAP4 expression in Jurkat (A), human liver (B) whole cell lysates. (Predicted band size: 31; 51 kD; Observed band size: 52 kD)

Immunofluorescent analysis of STEAP4 staining in Jurkat cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
