Background

Nuclear transport receptor that mediates nuclear import of proteins, such as histones, RPS3A, TNP2 and VDR . Serves as receptor for nuclear localization signals (NLS) in cargo substrates . Is thought to mediate docking of the importin/substrate complex to the nuclear pore complex (NPC) through binding to nucleoporin and the complex is subsequently translocated through the pore by an energy requiring, Ran-dependent mechanism . At the nucleoplasmic side of the NPC, Ran binds to the importin, the importin/substrate complex dissociates and importin is re-exported from the nucleus to the cytoplasm where GTP hydrolysis releases Ran .

Western blot analysis of RanBP4 expression in A549 (A), mouse lung (B), rat brain (C) whole cell lysates. (Predicted band size: 119 kD; Observed band size: 119 kD)

Immunofluorescent analysis of RanBP4 staining in L929 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a AREX® Fluor 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
