Background
The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes . Rabs cycle between an inactive GDP-bound form and an active GTP-bound form that is able to recruit to membranes different sets of downstream effectors directly responsible for vesicle formation, movement, tethering and fusion . RAB22A plays a role in endocytosis and intracellular protein transport. Mediates trafficking of transferrin/TF from early endosomes to recycling endosomes . Required for NGF-mediated endocytosis of NTRK1, and subsequent neurite outgrowth . Has low GTPase activity .

Western blot analysis of RAB22 expression in MCF7 (A), Hela (B), mouse liver (C), mouse kidney (D), rat liver (E), rat kidney (F) whole cell lysates. (Predicted band size: 21 kD; Observed band size: 22 kD)

Immunohistochemical analysis of RAB22 staining in human tonsil formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. Tyramide-AREX® Fluor 488 (green) was used as the chromogen. The section was then counterstained with DAPI (blue).

Immunofluorescent analysis of RAB22 staining in A375 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with an AREX® Fluor 488 -conjugated secondary antibody (green) in PBS at room temperature in the dark. Phalloidin - AREX® Fluor 594 was used to stain Actin filaments (red). DAPI was used to stain the cell nuclei (blue).
