Background

Thiol-specific peroxidase that catalyzes the reduction of hydrogen peroxide and organic hydroperoxides to water and alcohols, respectively . Can reduce H(2)O(2) and short chain organic, fatty acid, and phospholipid hydroperoxides . Also has phospholipase activity, can therefore either reduce the oxidized sn-2 fatty acyl group of phospholipids (peroxidase activity) or hydrolyze the sn-2 ester bond of phospholipids (phospholipase activity) . These activities are dependent on binding to phospholipids at acidic pH and to oxidized phospholipds at cytosolic pH . Plays a role in cell protection against oxidative stress by detoxifying peroxides and in phospholipid homeostasis .

Western blot analysis of Peroxiredoxin 6 expression in PC3 (A), K562 (B), mouse liver (C), mouse heart (D) whole cell lysates. (Predicted band size: 25 kD; Observed band size: 27 kD)

Immunofluorescent analysis of Peroxiredoxin 6 staining in K562 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.

Store at 4°C short term. For long term storage, store at -20°C, avoiding freeze/thaw cycles.
