Background

Serine protease inhibitor. Inhibits TMPRSS7 . Is a primary inhibitor of tissue-type plasminogen activator (PLAT) and urokinase-type plasminogen activator (PLAU). As PLAT inhibitor, it is required for fibrinolysis down-regulation and is responsible for the controlled degradation of blood clots . As PLAU inhibitor, it is involved in the regulation of cell adhesion and spreading . Acts as a regulator of cell migration, independently of its role as protease inhibitor . It is required for stimulation of keratinocyte migration during cutaneous injury repair . It is involved in cellular and replicative senescence . Plays a role in alveolar type 2 cells senescence in the lung .

Western blot analysis of PAI1 expression in A549 (A) whole cell lysates. (Predicted band size: 43; 45 kD; Observed band size: 45 kD)

Immunofluorescent analysis of PAI1 staining in human liver cancer tissues. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a Alexa Fluor 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.
