Background

Serine/threonine kinase that plays a role in the response to environmental stress and cytokines such as TNF. Appears to act upstream of the JUN N-terminal pathway . Activator of the Hippo signaling pathway which plays a pivotal role in organ size control and tumor suppression by restricting proliferation and promoting apoptosis. MAP4Ks act in parallel to and are partially redundant with STK3/MST2 and STK4/MST2 in the phosphorylation and activation of LATS1/2, and establish MAP4Ks as components of the expanded Hippo pathway . Phosphorylates SMAD1 on Thr-322 .

Western blot analysis of NGK expression in DLD (A) whole cell lysates. (Predicted band size: 142 kD; Observed band size: 150 kD)

Immunofluorescent analysis of NGK staining in A549 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a Alexa Fluor 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.

Store at 4°C short term. For long term storage, store at -20°C, avoiding freeze/thaw cycles.
