Background

Inhibits the activity of dimeric NF-kappa-B/REL complexes by trapping REL (RELA/p65 and NFKB1/p50) dimers in the cytoplasm by masking their nuclear localization signals dimers in the cytoplasm by masking their nuclear localization signals . On cellular stimulation by immune and pro-inflammatory responses, becomes phosphorylated promoting ubiquitination and degradation, enabling the dimeric RELA to translocate to the nucleus and activate transcription .

Western blot analysis of IKB alpha (Phospho-Y42) expression in K562 (A), Jurkat IL1b-treated (B), Raw264.7 LPS-treated (C), H9C2 LPS-treated (D) whole cell lysates. (Predicted band size: 35 kD; Observed band size: 37 kD)

Immunohistochemical analysis of IKB alpha (Phospho-Y42) staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
