Background

Core component of nucleosome. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling.

Western blot analysis of Histone H4 (Phospho-S47) expression in HEK293T (A), U2OS (B) whole cell lysates. (Predicted band size: 11 kD; Observed band size: 11 kD)

Immunofluorescent analysis of Histone H4 (Phospho-S47) staining in HeLa cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.

Storage

Store at 4°C short term. For long term storage, store at -20°C, avoiding freeze/thaw cycles.
