Background

Core component of multiple cullin-RING-based E3 ubiquitin-protein ligase complexes which mediate the ubiquitination of target proteins . As a scaffold protein may contribute to catalysis through positioning of the substrate and the ubiquitin-conjugating enzyme . The E3 ubiquitin-protein ligase activity of the complex is dependent on the neddylation of the cullin subunit and is inhibited by the association of the deneddylated cullin subunit with TIP120A/CAND1 . The functional specificity of the E3 ubiquitin-protein ligase complex depends on the variable substrate recognition component . DCX(DET1-COP1) directs ubiquitination of JUN . DCX(DDB2) directs ubiquitination of XPC .

Western blot analysis of Cullin 4A expression in Hela (A), HepG2 (B), mouse brain (C) whole cell lysates. (Predicted band size: 76; 87 kD; Observed band size: 88 kD)

Immunofluorescent analysis of Cullin 4A staining in U2OS cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.
