Background
Cell adhesion molecule which is required for leukocyte transendothelial migration (TEM) under most inflammatory conditions under most inflammatory conditions . Tyr-690 plays a critical role in TEM and is required for efficient trafficking of PECAM1 to and from the lateral border recycling compartment (LBRC) and is also essential for the LBRC membrane to be targeted around migrating leukocytes . Trans-homophilic interaction may play a role in endothelial cell-cell adhesion via cell junctions . Heterophilic interaction with CD177 plays a role in transendothelial migration of neutrophils . Homophilic ligation of PECAM1 prevents macrophage-mediated phagocytosis of neighboring viable leukocytes by transmitting a detachment signal .

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human placenta tissue labelling CD31 with ARA812 at 1:3,200. Heat mediated antigen retrieval was performed using Tris/EDTA buffer pH 9.0.

Overlay histogram showing Jurkat cells stained with ARA812 (Red). The cells were fixed with 4% paraformaldehyde for 10 min. The cells were then incubated in the antibody (ARA812, 1:1,000 dilution) in 1x PBS/1% BSA for 30 min at room temperature. The secondary antibody used was a Goat Anti-Rabbit Alexa Fluor<sup>®</sup> 488 (IgG H+L) at 1:2,000 dilution for 20 min at room temperature. Unlabelled sample (Black) was used as a control.

CD31 was immunoprecipitated from 0.4mg of THP-1 whole cell lysate with ARA812 at 1:50 dilution.
