Background

Promotes cell death. Successfully competes for the binding to Bcl-X(L), Bcl-2 and Bcl-W, thereby affecting the level of heterodimerization of these proteins with BAX. Can reverse the death repressor activity of Bcl-X(L), but not that of Bcl-2 . Appears to act as a link between growth factor receptor signaling and the apoptotic pathways.

Western blot analysis of BAD expression in A375 (A), HEK293T (B), Hela (C), AML12 (D) whole cell lysates. (Predicted band size: 18 kD; Observed band size: 23 kD)

Immunohistochemical analysis of BAD staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
