Background

G-protein coupled receptor for 5-hydroxytryptamine (serotonin) . Also functions as a receptor for various ergot alkaloid derivatives and psychoactive substances . Ligand binding causes a conformation change that triggers signaling via guanine nucleotide-binding proteins (G proteins) and modulates the activity of downstream effectors . HTR2B is coupled to G(q)/G(11) G alpha proteins and activates phospholipase C-beta, releasing diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3) second messengers that modulate the activity of phosphatidylinositol 3-kinase and promote the release of Ca(2+) ions from intracellular stores, respectively . Beta-arrestin family members inhibit signaling via G proteins and mediate activation of alternative signaling pathways .

Western blot analysis of 5-HT2B expression in Hela (A), rat liver (B), rat heart (C), rat kidney (D) whole cell lysates. (Predicted band size: 54 kD; Observed band size: 54 kD)

Immunofluorescent analysis of 5-HT2B staining in COS7 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a AREX® Fluor 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.

Storage

Store at 4°C short term. For long term storage, store at -20°C, avoiding freeze/thaw cycles.
