Background

Non-selective calcium permeant cation channel involved in detection of noxious chemical and thermal stimuli . Seems to mediate proton influx and may be involved in intracellular acidosis in nociceptive neurons. Involved in mediation of inflammatory pain and hyperalgesia. Sensitized by a phosphatidylinositol second messenger system activated by receptor tyrosine kinases, which involves PKC isozymes and PCL. Activated by vanilloids, like capsaicin, and temperatures higher than 42 degrees Celsius . Upon activation, exhibits a time- and Ca(2+)-dependent outward rectification, followed by a long-lasting refractory state. Mild extracellular acidic pH (6.5) potentiates channel activation by noxious heat and vanilloids, whereas acidic conditions (pH <6) directly activate the channel.

Western blot analysis of TRPV1 expression in U87MG (A), A375 (B) whole cell lysates. (Predicted band size: 94 kD; Observed band size: 110 kD)

Immunofluorescent analysis of TRPV1 staining in Hela cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a AREX® Fluor 488 -conjugated secondary antibody (green) in PBS at room temperature in the dark. Phalloidin - AREX® Fluor 555 was used to stain Actin filaments (red). DAPI was used to stain the cell nuclei (blue).

Storage

Store at 4°C short term. For long term storage, store at -20°C, avoiding freeze/thaw cycles.
