Background

Protein kinase involved in intracellular signaling pathways downstream of integrins and receptor-type kinases that plays an important role in cytoskeleton dynamics, in cell adhesion, migration, proliferation, apoptosis, mitosis, and in vesicle-mediated transport processes . Can directly phosphorylate BAD and protects cells against apoptosis . Activated by interaction with CDC42 and RAC1 . Functions as a GTPase effector that links the Rho-related GTPases CDC42 and RAC1 to the JNK MAP kinase pathway . Phosphorylates and activates MAP2K1, and thereby mediates activation of downstream MAP kinases . Involved in the reorganization of the actin cytoskeleton, actin stress fibers and of focal adhesion complexes .

Western blot analysis of PAK1/2/3 (Phospho-S144/141/139) expression in HepG2 (A), mouse brain (B) whole cell lysates. (Predicted band size: 60; 58; 62 kD; Observed band size: 68 kD)

Immunohistochemical analysis of PAK1/2/3 (Phospho-S144/141/139) staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

Store at 4°C short term. For long term storage, store at -20°C, avoiding freeze/thaw cycles.
