Background

Cellular uptake of iron occurs via receptor-mediated endocytosis of ligand-occupied transferrin receptor into specialized endosomes . Endosomal acidification leads to iron release. The apotransferrin-receptor complex is then recycled to the cell surface with a return to neutral pH and the concomitant loss of affinity of apotransferrin for its receptor. Transferrin receptor is necessary for development of erythrocytes and the nervous system . A second ligand, the hereditary hemochromatosis protein HFE, competes for binding with transferrin for an overlapping C-terminal binding site. Positively regulates T and B cell proliferation through iron uptake . Acts as a lipid sensor that regulates mitochondrial fusion by regulating activation of the JNK pathway .

Western blot analysis of CD71 expression in mouse kidney (A), rat liver (B), rat kidney (C) whole cell lysates. (Predicted band size: 84 kD; Observed band size: 90 kD)

Immunohistochemical analysis of CD71 staining in rat liver formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. Tyramide-AREX® Fluor 488 (green) was used as the chromogen. The section was then counterstained with DAPI (blue).
