Background

Critical isomerohydrolase in the retinoid cycle involved in regeneration of 11-cis-retinal, the chromophore of rod and cone opsins. Catalyzes the cleavage and isomerization of all-trans-retinyl fatty acid esters to 11-cis-retinol which is further oxidized by 11-cis retinol dehydrogenase to 11-cis-retinal for use as visual chromophore . Essential for the production of 11-cis retinal for both rod and cone photoreceptors . Also capable of catalyzing the isomerization of lutein to meso-zeaxanthin an eye-specific carotenoid . The soluble form binds vitamin A (all-trans-retinol), making it available for LRAT processing to all-trans-retinyl ester.

Western blot analysis of RPE65 expression in Hela (A), mouse heart (B), rat brain (C) whole cell lysates. (Predicted band size: 60 kD; Observed band size: 61 kD)

Immunofluorescent analysis of RPE65 staining in rat eye. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a AREX® Fluor 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
