Mediates sumoylation of ALKBH5, AXIN1, CEBPA, KLF8, GATA2, PARK7, HERC2, MYB, TCF4 and RNF168 . Plays a crucial role as a transcriptional coregulation in various cellular pathways, including the STAT pathway, the p53/TP53 pathway, the Wnt pathway and the steroid hormone signaling pathway . Involved in gene silencing . In Wnt signaling, represses LEF1 and enhances TCF4 transcriptional activities through promoting their sumoylations . Enhances the sumoylation of MTA1 and may participate in its paralog-selective sumoylation .

Western blot analysis of PIAS4 expression in HepG2 (A), HeLa (B) whole cell lysates. (Predicted band size: 56 kD; Observed band size: 75 kD)

Immunohistochemical analysis of PIAS4 staining in human brain formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
