Background

DNA glycosylase which prefers single-stranded DNA (ssDNA), or partially ssDNA structures such as bubble and fork structures, to double-stranded DNA (dsDNA) , or partially ssDNA structures such as bubble and fork structures, to double-stranded DNA (dsDNA) . Mediates interstrand cross-link repair in response to replication stress: acts by mediating DNA glycosylase activity, cleaving one of the two N-glycosyl bonds comprising the interstrand cross-link, which avoids the formation of a double-strand break but generates an abasic site that is bypassed by translesion synthesis polymerases .

Western blot analysis of NEIL3 expression in MCF7 (A), H1792 (B), rat thymus (C), mouse testis (D) whole cell lysates. (Predicted band size: 67 kD; Observed band size: 68 kD)

Immunohistochemical analysis of NEIL3 staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
