Background

Produces nitric oxide (NO) which is a messenger molecule with diverse functions throughout the body which is a messenger molecule with diverse functions throughout the body . In macrophages, NO mediates tumoricidal and bactericidal actions. Also has nitrosylase activity and mediates cysteine S-nitrosylation of cytoplasmic target proteins such PTGS2/COX2 . As component of the iNOS-S100A8/9 transnitrosylase complex involved in the selective inflammatory stimulus-dependent S-nitrosylation of GAPDH on 'Cys-247' implicated in regulation of the GAIT complex activity and probably multiple targets including ANXA5, EZR, MSN and VIM . Involved in inflammation, enhances the synthesis of pro-inflammatory mediators such as IL6 and IL8 .

Western blot analysis of iNOS (Phospho-Y151) expression in HeLa (A), Jurkat (B), MCF7 (C) whole cell lysates. (Predicted band size: 131 kD; Observed band size: 130 kD)

Immunohistochemical analysis of iNOS (Phospho-Y151) staining in human colon cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

Direct ELISA antibody dose-response curve using Anti-iNOS (Phospho-Y151) Antibody. Antigen (Phosphopeptide and non-phosphopeptide) concentration is 5 ug/ml. Goat Anti-Rabbit IgG (H&L) - HRP was used as the secondary antibody, and signal was developed by TMB substrate.

Store at 4°C short term. For long term storage, store at -20°C, avoiding freeze/thaw cycles.
