Background
The Exosome Protein Lysis Buffer is a specialized reagent developed by Shanghai Umibio Biotechnology Co., Ltd. for exosome samples. It efficiently lyses exosome proteins and contains protease inhibitors to effectively prevent protein degradation. To use, simply mix 1:1 with the exosome sample. The lysed protein samples can be used for BCA, Western Blot, and other experiments. This reagent contains a high concentration of detergents and is not suitable for protein concentration determination by the Bradford method.
The main components of this product include Tris (pH 7.4), NaCl, Triton X-100, sodium deoxycholate, SDS, EDTA, PMSF, and a Protease Inhibitor Cocktail (AEBSF, Aprotinin, Bestatin, E-64, Leupeptin, Pepstatin A), among others.

Product Usage
1.Preparation: Thaw the lysis buffer at 2~8°C, mix well, and place on ice before use. 2.Lysis: Mix the exosome sample with the lysis buffer at a 1:1 volume ratio and incubate on ice for 10 minutes. 3.Centrifugation: Centrifuge at 11,200 ×g (~12,000 rpm*) at 4°C for 5 minutes. Collect the supernatant for protein concentration determination using the BCA method and subsequent experiments such as Western Blot. Calculated for a small centrifuge with an effective radius of approximately 7 cm (≤ 2 mL centrifuge tubes).
