Background

Histone H3-like nucleosomal protein that is specifically found in centromeric nucleosomes . Replaces conventional H3 in the nucleosome core of centromeric chromatin that serves as an assembly site for the inner kinetochore . The presence of CENPA subtly modifies the nucleosome structure and the way DNA is wrapped around the nucleosome and gives rise to protruding DNA ends that are less well-ordered and rigid compared to nucleosomes containing histone H3 . May serve as an epigenetic mark that propagates centromere identity through replication and cell division . Required for recruitment and assembly of kinetochore proteins, and as a consequence required for progress through mitosis, chromosome segregation and cytokinesis .

Western blot analysis of CENPA (Phospho-S7) expression in HEK293T (A), Hela (B), DLD (C), rat testis (D), rat spleen (E) whole cell lysates. (Predicted band size: 15 kD; Observed band size: 17 kD)

Immunofluorescent analysis of CENPA (Phospho-S7) staining in HeLa cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a Alexa Fluor 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.

Direct ELISA antibody dose-response curve using Anti-CENPA (Phospho-S7) Antibody. Antigen (Phosphopeptide and non-phosphopeptide) concentration is 5 ug/ml. Goat Anti-Rabbit IgG (H&L) - HRP was used as the secondary antibody, and signal was developed by TMB substrate.
