Background
5-HT7 Receptor is a Serotonin Receptor with several functions, including regulation of the circadian rhythm and mediation of smooth-muscle relaxation in the vasculature and GI tract. This receptor has been implicated in depression, autism, and other neuropsychiatric disorders. Three protein isoforms have been documented: isoform a: 445aa; isoform b: 432aa; isoform d: 479aa. 5-HT7 Receptor has been reported in aorta, brain, heart, placenta, small intestine, spleen, artery, and vessel. An EST has been isolated from a testis library.

Western blot analysis of 5-HT7 expression in Hela (A), H1688 (B) whole cell lysates. (Predicted band size: 53 kD; Observed band size: 54 kD)

Immunofluorescent analysis of 5-HT7 staining in COS7 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a AF594-conjugated secondary antibody (red) in PBS at room temperature in the dark.
