Background

Non-selective calcium permeant cation channel . It is activated by innocuous (warm) temperatures and shows an increased response at noxious temperatures greater than 39 degrees Celsius . Activation exhibits an outward rectification . The channel pore can dilate to provide permeability to larger cations . May associate with TRPV1 and may modulate its activity . Is a negative regulator of hair growth and cycling: TRPV3-coupled signaling suppresses keratinocyte proliferation in hair follicles and induces apoptosis and premature hair follicle regression (catagen) .

Western blot analysis of TRPV3 expression in U87MG (A), MCF7 (B), C6 (C), CT26 (D) whole cell lysates. (Predicted band size: 90 kD; Observed band size: 100 kD)

Immunohistochemical analysis of TRPV3 staining in human colon cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
