Background

Precursor of the transcription factor form (Processed sterol regulatory element-binding protein 1), which is embedded in the endoplasmic reticulum membrane , which is embedded in the endoplasmic reticulum membrane . Low sterol concentrations promote processing of this form, releasing the transcription factor form that translocates into the nucleus and activates transcription of genes involved in cholesterol biosynthesis and lipid homeostasis .

Western blot analysis of SREBP1 (Phospho-S439) expression in H460 (A) whole cell lysates. (Predicted band size: 121 kD; Observed band size: 125; 68 kD)

Immunohistochemical analysis of SREBP1 (Phospho-S439) staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
