Background

Phosphorylates serines and threonines, but also appears to possess tyrosine kinase activity . Involved in DNA damage checkpoint control and for proper DNA damage repair . In response to injury that includes DNA damage, NEK1 phosphorylates VDAC1 to limit mitochondrial cell death . May be implicated in the control of meiosis . Involved in cilium assembly .

Western blot analysis of NEK1 expression in human fetal kidney (A) whole cell lysates. (Predicted band size: 142 kD; Observed band size: 143 kD)

Immunohistochemical analysis of NEK1 staining in human hepatocarcinoma formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
