Background

Component of the integrator complex, a multiprotein complex that terminates RNA polymerase II (Pol II) transcription in the promoter-proximal region of genes transcription in the promoter-proximal region of genes . The integrator complex provides a quality checkpoint during transcription elongation by driving premature transcription termination of transcripts that are unfavorably configured for transcriptional elongation: the complex terminates transcription by (1) catalyzing dephosphorylation of the C-terminal domain (CTD) of Pol II subunit POLR2A/RPB1 and SUPT5H/SPT5, (2) degrading the exiting nascent RNA transcript via endonuclease activity and (3) promoting the release of Pol II from bound DNA .

Western blot analysis of INTS3 expression in 293 (A), Hela (B), Jurkat (C), NIH3T3 (D) whole cell lysates. (Predicted band size: 118 kD; Observed band size: 118 kD)

Flow cytometric analysis of Hela cells using Anti-INTS3 Antibody. The cells were fixed with 2% paraformaldehyde (10 min) and then permeabilized with 90% methanol for 10 min. The cells were incubated in 2% bovine serum albumin to block non-specific protein-protein interactions followed by the antibody at 37 °C for 60 min. The secondary antibody Goat Anti-Rabbit IgG (H&L) - AREX® Fluor 488 was incubated at 37 °C for 40 min. Isotype control antibody (blue line) was used under the same condition.
