Background

Interferon (IFN)-inducible GTPase that plays important roles in innate immunity against a diverse range of bacterial, viral and protozoan pathogens -inducible GTPase that plays important roles in innate immunity against a diverse range of bacterial, viral and protozoan pathogens . Hydrolyzes GTP to GMP in 2 consecutive cleavage reactions, but the major reaction product is GDP . Following infection, recruited to the pathogen-containing vacuoles or vacuole-escaped bacteria and acts as a positive regulator of inflammasome assembly by promoting the release of inflammasome ligands from bacteria . Acts by promoting lysis of pathogen-containing vacuoles, releasing pathogens into the cytosol .

Data

Western blot analysis of GBP2 expression in A375 (A) whole cell lysates. (Predicted band size: 67 kD; Observed band size: 67 kD)

Immunofluorescent analysis of GBP2 staining in L929 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a AREX® Fluor 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
