Background

DNA methyltransferase that methylates CpG residues . Preferentially methylates hemimethylated DNA . Associates with DNA replication sites in S phase maintaining the methylation pattern in the newly synthesized strand, that is essential for epigenetic inheritance . Associates with chromatin during G2 and M phases to maintain DNA methylation independently of replication . It is responsible for maintaining methylation patterns established in development . DNA methylation is coordinated with methylation of histones . Mediates transcriptional repression by direct binding to HDAC2 . In association with DNMT3B and via the recruitment of CTCFL/BORIS, involved in activation of BAG1 gene expression by modulating dimethylation of promoter histone H3 at H3K4 and H3K9 .

Western blot analysis of DNMT1 expression in SW480 (A) whole cell lysates. (Predicted band size: 144; 183; 184 kD; Observed band size: 200 kD)

Immunofluorescent analysis of DNMT1 staining in A549 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.

Store at 4°C short term. For long term storage, store at -20°C, avoiding freeze/thaw cycles.
